    • MR-VP Medium, also known as Glucose Phosphate Broth or Buffered Glucose Broth, is used for the differentiation of the coli-aerogenes group of bacteria.
    • It specifically facilitates the Methyl Red (MR) and Voges-Proskauer (VP) reactions.
  • Components:
    • Contains glucose, peptones, and phosphate buffer.
    • Adjusted to meet performance standards.
  • Methyl Red (MR) Test:
    • Determines the hydrogen ion concentration (pH) when an organism ferments glucose.
    • Initially, all enteric bacteria are MR-positive (red color).
    • Over time, MR-positive organisms continue to produce acids, resulting in a low pH and sustained red color.
    • MR-negative organisms metabolize fermentation products differently, leading to a neutral pH.
  • Voges-Proskauer (VP) Test:
    • Detects acetylmethylcarbinol derived from glucose metabolism.
    • Primarily used to differentiate Escherichia coli (VP-negative) from the Klebsiella-Enterobacter group (VP-positive).
    • Acetylmethylcarbinol is a precursor to 2,3-butanediol production.
    • VP-positive organisms produce a pink-red color due to oxidation of acetylmethylcarbinol.
  • Preparation:
    • Suspend 17 grams of dehydrated medium in 1000 ml of demineralized water.
    • Warm if necessary to dissolve.
    • Incubate inoculated MR-VP Broth for a minimum of 48 hours (up to 5 days for MR test and up to 10 days for VP test).

      • Dichloran inhibits spreading of mucoraceous fungi and restricts colony size.
    • Application:
      • Used for counting yeasts and molds in foodstuffs.
      • Suitable for dried fruits, spices, confectionery, cereals, nuts, dried meat, and fish products.
    • Selective Properties:
